cAMP radioimmunoassay without interference from calcium or EDTA.

چکیده
Calcium and EDTA interfere with radioimmunoassay of cAMP or acetylated cAMP. Both interferences with the assay of acetylated cAMP can be obviated by including in the buffer 15 mmol of CaCl2 per liter, which at the same time increases the affinity of the antibody by 89%. As a result, antibody can be decreased by 65%. This method requires no prepurification (extraction or chromatography), and no correction for analytical recovery is necessary. This assay can detect 12 fmol of cAMP per tube, so only microliter quantities of plasma are needed.

Positive interference in Radioimmunoassay determination of serum total T3
Introduction: Determination of thyrotropin, total and free thyroxine and triiodothyronine are widely used as diagnostic methods for thyroid function evaluation. There have been numerous reports of interferences in thyroid hormone immunoassays. Herein, a prominent positive interference is reported for a radioimmunoassay kit of total triiodothyronine. Materials and Methods: In this study, a total...
متن کاملDecreased intracellular calcium stimulates renin release via calcium-inhibitable adenylyl cyclase.
Intracellular calcium and cAMP are the 2 second messengers that regulate renin release; cAMP stimulates renin release from juxtaglomerular (JG) cells, whereas increased intracellular calcium inhibits it. We hypothesized that decreased intracellular calcium acts by activating calcium-inhibitable isoforms of adenylyl cyclase, increasing cAMP, and stimulating renin secretion. We used a primary cul...
متن کاملInvestigation and comparison of calcium aluminate phases obtained by three methods of sol-gel (with EDTA), sol-gel (without EDTA) and solid-phase synthesis
In this study, we investigate calcium aluminate phases that were prepared by three different solid phase synthesis methods and a sol-gel method (with and without EDTA). The purpose of this study is to compare these three methods in terms of achieving higher purity, optimal phase and complexity in the production of the final product. Comparing the results, we found that the sol-gel method is mo...
متن کاملAssociated disturbances in calcium homeostasis and G protein-mediated cAMP signaling in bipolar I disorder.
BACKGROUND Evidence of extensive cross-talk between calcium (Ca(2+))- and cAMP-mediated signaling systems suggests that previously reported abnormalities in Ca(2+) homeostasis in bipolar I (BP-I) patients may be linked to disturbances in the function of G proteins that mediate cAMP signaling. METHODS To test this hypothesis, the beta-adrenergic agonist, isoproterenol, and the G protein activa...
متن کاملPectic Acid Effects on Prolactin Secretion in GH3/B6 Rat Pituitary Cell Line
Background: Pectic acid extracted from plants increases the secretion of prolactin (PRL) when injected intravenously into ewes or fed to rats. Fragments of ewe hypophysis and lactating rabbit mammary gland incubated in vitro in the presence of pectic acid secreted more PRL and caseins compared to the controls. However, it is not known whether pectic acid directly stimulates PRL secretion in pi...
